The saccharification was increased to 94.88% by removal of the hydrolysate after 24h by using a two-step hydrolysis. Significant lactic acid production (27.8mg/mL) was obtained by separate saccharification and fermentation after 72h of incubation.

Background: Simultaneous saccharification and fermentation (SSF) is a promising process for bioconversion of lignocellulosic biomass. High glucan loading for hydrolysis and fermentation is an efficient approach to reduce the capital costs for bio-based products production.

The bioconversion of cellulosic wastes into high-value bio-products by saccharification and fermentation processes is an important step that can reduce the environmental pollution caused by agricultural wastes. Saccharification-Fermentation process was developed. Saccharification and fermentation are done simultaneously in order to decrease enzymatic end-product inhibition thus producing a better yield. Such process involves the use of bioreactor for controlling parameters like temperature and pH where enzymes can perform its best. 2019-01-22 · The simultaneous saccharification and fermentation (SSF) process was conceptualized in the late 1970s by Wright et al., Takagi et al., and Blotkamp et al. [ 11, 12 ].

Saccharification and fermentation

Simultaneous saccharification and fermentation (SSF) and pre-hydrolysis with SSF (PSSF) were used to produce hydrogen from the biomass of Chlorella sp. SSF was conducted using an enzyme mixture consisting of 80 filter paper unit (FPU) g-biomass−1 of cellulase, 92 U g-biomass−1 of amylase, and 120 U g-biomass−1 of glucoamylase at 35 °C for 108 h.

These unit operations can be combined either sequen-tially or in a simultaneous saccharification–fermentation (SSF) process. For example, a fully consolidated high gravity process Simultaneous Saccharification and Fermentation (SSF) of Pretreated Cogongrass.

SSF was conducted using an enzyme mixture consisting of 80 filter paper unit (FPU) g-biomass−1 of cellulase, 92 U g-biomass−1 of amylase, and 120 U g-biomass−1 of glucoamylase at 35 °C for 108 h. Detoxification of the acid‐ and enzyme‐treated rice hull hydrolyzate by overliming (pH 10.5, 90 °C, 30 min) reduced the time required for maximum ethanol production (17 ± 0.2 g from 42.0 ± 0.7 g sugars per L) by the E. coli strain from 64 to 39 h in the case of separate hydrolysis and fermentation and increased the maximum ethanol yield (per L) from 7.1 ± 2.3 g in 140 h to 9.1 ± 0.7 g in 112 h in the case of simultaneous saccharification and fermentation.

This research shows the effect of dilute acid pretreatment with various sulfuric acid concentrations (0.5–2.0% [wt/vol]) on enzymatic saccharification and fermentation yield of rye straw. After pretreatment, solids of rye straw were suspended in Na citrate buffer or post‐pretreatment liquids (prehydrolysates) containing sugars liberated after hemicellulose hydrolysis.
